Sonophotodynamic therapy-a new method in the treatment of Cutaneous Leishmaniasis: An in vitro study

OBJECTİVE: In this study, we aimed to examine of antileishmanial effect curcumin-mediated sonodynamic (SDT), photodynamic (PDT), and sonophotodynamic (SPDT) therapies with a potential mechanism against the Leishmania tropica (L.tropica) promastigotes. SPDT is new treatment modality for Leishmaniasis that combines therapies. Curcumin natural antiinflammatory agent has been used treating medical conditions many years. was in study both as sonosensitizer photosensitizer compare PDT, SDT, on L.tropica promastigotes.
 MATERIALS AND METHODS: The cells were incubated different concentrations (0.25, 1,4,16 64μM) curcumin 1 hour, exposed 3 W/cm2 intensity ultrasound 1MHz frequency and/or subjected 1,32 J/cm2 light irradiation 30 minutes. Also, parasite only presence or absence curcumin. XTT evaluate cell viability giemsa staining determine morphological changes.
 RESULTS: With combination ultrasound, light, mediated light,L.tropica promastigote found be decreased compared control, ultrasound-control light-control group. greatest reduction Giemsa findings showed PDT induced several alterations promastigotes atypical. These results more effective than other promastigotes
